Abstract

The production of exo-polysaccharide (β-glucan and pullulan) by A. pullulans strain APX1.5.4.1 was affected by temperature, fermentation time and pH of fermentation medium. The aim of this study was to determine the fermentation conditions for high β-glucan production performed by response surface method (RSM). The results showed that the suitable fermentation conditions for high β-glucan production (11,215±0.083 g/L) were determined as fermentation temperature at 28oC, fermentation time 3 days and pH 6. In this fermentation condition, the obtained pullulan content is quite high (20.48±0.15 g/L). The content of β-glucan produced from strain A. pullulans APX1.5.4.1 in this study was 6.02 times higher than the amount of β-glucan (1,862 g/L) from strain A. pullulans KSY-0516 and 1.22 times higher than that of strain A. pullulans KSY-0516. with β-glucan (9.2 g/L) from A. pullulans M-2 strain. The use of response surface method helped determine the suitable fermentation conditions for β-glucan and pullulan production with R2 values of 0.9955 and 0.9960, respectively.
